Tiivistelmä
Plasman katekoliamiinit ovat monipuolinen tutkimuskohde (Ribeiro ym. 2016). Katekoliamiinien pitoisuuksien määritys biologisista näytteistä on edelleen analyyttisesti haasteellista, huolimatta useista menetelmistä, joita tähän tarkoitukseen on kehitelty vuosien varrella. Suurimmat haasteet plasman katekoliamiinien analysoinnissa johtuvat niiden taipumuksesta hapettua helposti ja niiden pienistä pitoisuuksista. (Bicker 2013.)
Tämän opinnäytetyön tavoitteena oli analysoida erään taustamelun stressivaikutuksia arvioivan tutkimushankkeen näytteistä plasman katekoliamiinien pitoisuudet HPLC-EC -menetelmällä luotettavasti ja laadukkaasti. Tämä edellytti menetelmän optimointia. Opinnäytetyön toimeksianto tuli Turun yliopiston Biolääketieteen laitokselta ja opinnäytetyö toteutettiin Biolääketieteen laitoksen Bioanalyyttisessa laboratoriossa. Bioanalyyttinen laboratorio vastasi opinnäytetyön toteutuksen ohjauksesta ja kuluista. Opinnäytetyön tuotoksena syntyi päivitetty menetelmäohje Bioanalyyttiselle laboratoriolle.
Tässä raportissa käsitellään plasman katekoliamiinien analysoinnin tietoperustaa ja esitellään opinnäytetyöprojektin toteutumista. Tietoperustassa käsitellään katekoliamiineja ja niiden analyyttista metodologiaa, erityisesti opinnäytetyöprojektissa käytettyä HPLC-EC-menetelmää ja alumiinioksidiuuttoa. Lisäksi raportissa kerrotaan toteutuneesta menetelmän optimoinnista ja syntyneestä tuotoksesta, menetelmäohjeesta.
Menetelmän optimoinnissa onnistuttiin parantamaan huomattavasti menetelmän herkkyyttä. Tulevaisuudessa voitaisiin vielä jatkaa työtä sopivan ajoliuoksen valmistamiseksi itse ja menetelmän herkkyyden parantamiseksi entisestään. Opinnäytetyön toimeksiantaja oli tyytyväinen syntyneeseen menetelmäohjeeseen ja uutta menetelmäohjetta hyödynnettiin tutkimushankkeen näytteiden analysoinnissa. Determination of catecholamines in plasma is needed for several purposes (Ribeiro et al. 2016). Despite the wide range of methods developed for this purpose, determination of plasma catecholamines is still an analytical challenge. The greatest issues stem from oxidation of catecholamines in the samples and the low levels of catecholamines in blood. (Bicker 2013.)
This thesis project aimed to determination of plasma catecholamines with high quality in a research project concerned with the stressor effects of background noise. In order to perform the analysis, the HPLC-EC method needed to be optimized. The request for this thesis project came from the Institute of Biomedicine of the University of Turku, and the project was actualized at the Bioanalytical Laboratory of the Institute of Biomedicine. As an outcome, the thesis project produced an updated method description for the determination of plasma catecholamines to the Bioanalytical Laboratory.
Catecholamines, the methods used for their determination in plasma and especially the method used in this research project, HPLC-EC with alumina extraction of the samples, are discussed in this thesis report. Also, the process of optimizing the method for research and the new method description are introduced.
The work to optimize the method significantly improved the method’s sensitivity. However, the expirements to improve the method’s sensitivity and to prepare the running buffer, instead of using commercial product, could be continued in the future projects. The new method description was approved by the Institute of Biomedicine and used in determination of plasma catecholamines for this research project.
