Tiivistelmä
Opinnäytetyö suoritettiin Turun ammattikorkeakoulussa, osana Uudet materiaalit ja prosessit-tutkimusryhmää sekä AMBioPharma-hanketta, jonka tavoitteena on kehittää uusia 3D-tulostettavia lääkeannosteluvalmisteita ja niiden valmistusprosesseja.
Tutkimuksen tavoitteena oli 3D-tulostettavan komposiittihydrogeelin, gelatiini-alginaatin, aikaansaaminen sekä tutkia onnistuneiden komposiittihydrogeelien reologisia ominaisuuksia reometrillä. Opinnäytetyössä testattiin EGFP:n kapselointia kolmella eri tavalla: EGFP:n kapselointi gelatiinikomponenttiin, EGFP:n kapselointi alginaattikomponenttiin ja EGFP:n kapselointi molempiin komponentteihin. Lisättävän EGFP:n määrä tutkittavaan komponenttiin oli 1 m-% hydrogeelin kokonaiskiintoaineen suhteen. Hydrogeelin liukenemista tutkittiin silmämääräisesti sekä hydrogeeliin kapseloidun EGFP:n vapautumisnopeutta komposiittihydrogeelistä dissoluutiotesteillä. EGFP-dissoluutiotestien tulosten analysointiin käytettävää fluoresenssimittausmenetelmää kehitettiin. Menetelmän kehitettävät asiat olivat puskuriliuoksen pH:n ja pinta-aktiivisen aineen vaikutus EGFP:n fluoresointiin ja niiden vaikutus mittaustulokseen sekä menetelmän mittausalueeseen.
Työssä saatiin kehitettyä kaksi eri valmistusprosessia 3D-tulostettavalle alginaatti-gelatiinihydrogeelille. Ristisilloitetun materiaalin rakenne oli käsiteltäessä vahvempi, mutta se pitää vielä varmistaa reologisilla mittauksilla. EGFP:n vapautumistutkimuksista saatiin eri materiaaleille vertailukelpoisia tuloksia, mutta kokonaisvapautuminen ei ollut 100 prosenttia, joten analyysimenetelmiä pitää vielä kehittää. The present Master’s thesis was completed at Turku University of Applied Sciences, as part of the New Materials and Processes research group and the AMBioPharma project, which aims to develop new 3D printable drug delivery products and their manufacturing processes.
The aim of the study is to obtain a 3D printable composite hydrogel, gelatin-alginate and to investigate the rheological properties of successful composite hydrogels with a rheometer. In the study, the encapsulation of EGFP was tested in three different ways: encapsulation of EGFP in the gelatin component, encapsulation of EGFP in the alginate component, and encapsulation of EGFP in both components. The amount of EGFP added to the test component was 1% by weight relative to the total solids of the hydrogel. The dissolution of the hydrogel was examined visually, and the rate of release of EGFP encapsulated in the hydrogel from the composite hydrogel by dissolution tests. A fluorescence measurement method for analyzing the results of the EGFP dissolution tests was developed. The issues to be developed in the method were the effect of the pH of the buffer solution and the effect of the surfactant on the fluorescence of EGFP and their effect on the measurement result as well as the measurement range of the method.
Two different manufacturing processes for 3D printable alginate-gelatin hydrogel were developed. The structure of the cross-linked material was stronger during processing, but this still needs to be confirmed by rheological measurements. EGFP release studies showed comparable results for different materials, but the overall release rate was not 100 percent, hence analytical methods still need to be developed.
